Here, we describe a multiple test matrix and
in vitro approach to assess the biological impact of differing e-cigarette activation mechanism (button vs puff-activated) and heating technology (cotton vs ceramic wick). The e-liquids selected for each device contained the same nicotine concentration and flavourings. We tested both e-liquid and aqueous extract of e-liquid aerosol using a high throughput
cytotoxicity and genotoxicity screen. We also conducted whole aerosol assessment both in a reconstituted human airway lung tissue (MucilAir) with associated endpoint assessment
(cytotoxicity, TEER, cilia beat frequency and active area) and an Ames whole aerosol assay with up to 900 consecutive undiluted puffs. Following this testing it is shown that the
biological impact of these devices is similar, taking into consideration the limitations and capturing efficiencies of the different testing matrices. We have contextualised these
responses against previous published reference cigarette data to establish the comparative reduction in response consistent with reduced risk potential of the e-cigarette products

Electronic cigarettes (e-cigarettes) are often considered a “safe substitute” for conventional cigarette cessation. The composition of the fluid is not always clearly defined and shows a large variation within brands and manufacturers. More than 80 compounds were detected in liquids and aerosols. E-cigarettes contain nicotine, and the addition of flavorings increases the toxicity of e-cigarette vapour in a significant manner. The heat generated by the e-cigarette leads to the oxidation
and decomposition of its components, eventually forming harmful constituents in the inhaled vapour. The effects of these toxicants on male and female reproduction are well established in conventional cigarette smokers. Although toxins were measured at much lower levels in e-cigarette aerosols compared to smoke from a conventional cigarette, there are concerns about their potential impact on male and female reproduction. The information available was mainly obtained from studies conducted in animal models, and investigations in humans are scarce. However, the effects observed in animal models suggest that caution should be taken when vaping and that more research needs to be conducted to identify its potential adverse effects on fertility. The prevalence of e-cigarette usage is alarming, and warnings should be made about the impact of vaping on reproductive health. To assess levels of nicotine, nicotine degradation products and some specific impurities in commercial refill liquids for electronic cigarettes.
Design and setting
We analyzed 20 models of 10 of the most popular brands of refill liquids, using gas and liquid chromatography.
Measurements
We assessed nicotine content, content of the known nicotine degradation products and impurities, and presence of ethylene glycol and diethylene glycol.
Findings
The nicotine content in the bottles corresponded closely to the labels on the bottles. The levels of nicotine degradation products represented 0–4.4% of those for nicotine, but for most samples the level was 1–2%. Cis-N-oxide, trans-N-oxide, myosmine, anatabine and anabasine were the most common additional compounds found. Neither ethylene glycol nor diethylene glycol were detected.
Conclusion
The nicotine content of electronic cigarette refill bottles is close to what is stated on the label. The plethora of product devices, formulations, and flavors, combined with the lack of industry standards and labeling requirements, quality control, and limited product oversight, has given rise to public concern about initiation of use and potential for adverse exposure and negative long-term health outcomes. This volume discusses how analytical methods can address these issues and support the manufacturing, labeling, distribution, testing, regulation, and monitoring for consistency of products with known chemical content and demonstrated performance characteristics.
The book begins with the background on aerosol drug delivery services and e-cigarettes, constituents of nicotine-containing liquid dosing formulations, typical use scenarios and associated aerosol emissions, and chemical exposures and pharmacological and toxicological effect profiles, and then continues with descriptions of the analytical methods used to characterize the chemicals in formulations and emissions from e-cigarettes, including their stability, physical particle-size distribution and thermal degradation under commonly employed conditions of use.
Analytical methods enabling detection of biomarkers of exposure and harm in complex biological matrices are discussed, with an emphasis on constituents or emissions of current medicinal interest or with potential to produce harm. However, little is known about the impact of different combinations of protonating acid on nicotine pharmacokinetics. The objectives of this study were to compare pharmacokinetics of nicotine absorption following use of a closed-system e-cigarette, containing e-liquids with two different nicotine levels and with different ratios of three common protonating acids—lactic, benzoic and levulinic. In a randomised, controlled, crossover study, nicotine pharmacokinetics and product liking were assessed for prototype e-liquids used in a Vuse e-cigarette containing either 3.5% or 5% nicotine and varying ratios of lactic, benzoic and/or levulinic acid. During an 8-day confinement period, 32 healthy adult current cigarette smokers/e-cigarette dual users used a single study e-liquid each day during 10-min fixed and ad libitum use periods after overnight nicotine abstinence. For most comparisons, Cmax and AUC 0–60 following both fixed and ad libitum puffing were significantly higher for e-liquids containing 5% nicotine compared with 3.5% nicotine. However, Cmax and AUC 0–60 were not statistically different for 5% nicotine e-liquids containing varying ratios of lactic, levulinic and benzoic acid when compared to an e-liquid containing lactic acid only. Mean scores for product liking were similar for all e-liquid formulations assessed, regardless of nicotine concentration, acid content, and whether the product was used in a fixed or ad libitum puffing regimen. While e-liquid nicotine concentration significantly affected users’ nicotine uptake, the different combinations of benzoic, levulinic and lactic acid in the e-liquids assessed had limited impact on nicotine pharmacokinetics and product liking scores. Most US health organizations, media coverage, and policymakers have focused primarily on risks to youths. Because of their messaging, much of the public—including most smokers—now consider e-cigarette use as dangerous as or more dangerous than smoking. By contrast, the National Academies of Science, Engineering, and Medicine concluded that e-cigarette use is likely far less hazardous than smoking. Policies intended to reduce adolescent vaping may also reduce adult smokers’ use of e-cigarettes in quit attempts. Les données récoltées à San Francisco, où une interdiction de vente de liquides de vapotage aromatisés est en vigueur depuis 2018, montrent pourtant une recrudescence significative du nombre de jeunes ayant fumé la cigarette après l’instauration de cette mesure, ce qui soulève de sérieux doutes sur l’efficacité de cette approche.
De plus, l’interdiction des saveurs de vapotage va priver plusieurs milliers de fumeurs adultes du meilleur outil disponible pour cesser de fumer, tel que documenté par plusieurs études cliniques récentes.
Le projet d’éliminer du marché les saveurs de vapotage semble donc mal avisé et nous croyons que son application devrait être à tout le moins retardée en attendant de mieux déterminer ses impacts sur les taux de tabagisme, autant chez les jeunes que chez les adultes.

a broad range of different electronic devices with which to inhale aerosols. The e-liquid vaporisation process comprises a chain of several chemical reactions which result in hundreds of chemical compounds. The composition and concentration of chemicals contained in e-liquids or, more precisely, in the resulting aerosol,are difficult to identify and quantify. This review article includes a summary of current evidence about the toxicity of selected chemical substances contained in
electronic cigarette liquids. Forty initial sources were selected in online databases on the basis of a nonsystematic
literature review using a set of keywords.
This overview presents selected categories of e-liquid ingredients according to their functions and chemical composition. The most frequent substances include bulking agents (propylene glycol and glycerol). So far,the least-explored category consists of a wide range of flavourings. A separate category is represented by nicotine, the cause of the development of physical dependence. Another, highly diverse group of compounds is chemical substances formed as byproducts of vaporisation. To review the available evidence evaluating the chemicals in refill solutions, cartridges, aerosols and environmental emissions of electronic cigarettes (e-cigarettes).
Methods
Systematic literature searches were conducted to identify research related to e-cigarettes and chemistry using 5 reference databases and 11 search terms. The search date range was January 2
007 to September 2013. The search yielded 36 articles, of which 29 were deemed relevant for analysis.
Results
The levels of nicotine, tobacco-specific nitrosamines (TSNAs), aldehydes, metals, volatile organic compounds (VOCs), flavours, solvent carriers and tobacco alkaloids in e-cigarette refill solutions, cartridges, aerosols and environmental emissions vary considerably. The delivery of nicotine and the release of TSNAs, aldehydes and metals are not consistent across products. Furthermore, the nicotine level listed on the labels of e-cigarette cartridges and refill solutions is often significantly different from measured values. Phenolic compounds, polycyclic aromatic hydrocarbons and drugs have also been reported in e-cigarette refill solutions, cartridges and aerosols. Varying results in particle size distributions of particular matter emissions from e-cigarettes across studies have been observed. Methods applied for the generation and chemical analyses of aerosols differ across studies. Performance characteristics of e-cigarette devices also vary across and within brands.
Conclusions Additional studies based on knowledge of e-cigarette user behaviours and scientifically validated aerosol generation and chemical analysis methods would be helpful in generating reliable measures of chemical quantities. This would allow comparisons of e-cigarette aerosol and traditional smoke constituent levels and would inform an evaluation of the toxicity potential of e-cigarettes

However, there is a major knowledge gap surrounding the effects of e-cig aerosols on pregnancy and fetal development. We aimed to evaluate the effects of vaping e-cigs during gestation on offspring growth and to asses if growth deficits are accompanied by altered maternal and fetal vascular hemodynamics. Sprague ?Dawley dams were assigned to Pair-Fed Control, Pair-Fed Juice, or Juice+Nicotine groups, and then underwent either a prenatal or prenatal+postnatal exposure paradigm in a custom-engineered vaping system. Mass spectrometry identified major aerosolized constituents from e-cig vaping. The Juice+Nicotine group exhibited significantly decreased fetal weight and crown-rump length (#46.56%, and #23.83%, respectively). Pre- and postnatal exposure to Juice+Nicotine resulted in decreased pup weight at postnatal day (PND) 4?10. Crown-rump length was decreased by 24.71% on PND 10. Blood flow in the Juice+Nicotine group was decreased in the maternal uterine and fetal umbilical circuits by 49.50% and 65.33%, respectively. We conclude that chronic exposure to e-cig aerosols containing nicotine during early development can have deleterious health effects on the exposed offspring. Vaping e-cigs containing nicotine during pregnancy lead to a reduction in offspring weight and crown-rump length, associated with a marked decrease in blood flow in both the maternal uterine and fetal umbilical circulation (a strong indicator of growth restriction). Individuals experienced increased social isolation resulting from the COVID-19 pandemic. Studies have found social isolation and loneliness to be strongly associated with anxiety and depression, which have been associated with increased smoking and vaping rates among young
adults, including college students.
Objectives:
To examine relationships between psychological distress and nicotine use within the context of the COVID-19 pandemic.
Methods:
A cross-sectional online survey (n = 4634; 77.9% female) was used to collect nicotine use and psychological measures from students enrolled at a large Midwestern university. Timeline follow-back data were collected from students reporting current cigarette or electronic cigarette (e-cigarette) use in the week before and immediately following the closure of campus due to the pandemic. Generalized estimating equations were used to examine the interaction between nicotine use and psychological symptoms across the 2-week period.
Results:
Both cigarette (Rate ratio (RR) = 1.115, 95% CI = 1.061, 1.171, p < .0001) and e-cigarette (β = 0.258, 95% CI = 0.166, 0.351, p < .0001) use increased significantly following campus closure. Students experiencing higher levels of depression reported greater increases in e-cigarette use frequency over time as compared to students reporting fewer symptoms of depression (β = 0.018, 95% CI = 0.006, 0.030, p = .004).
Conclusions: Increases in nicotine use were found immediately following the implementation of public health safety measures that closed most university campuses. Additional and/or increased stressors have potentially impacted young adults who are college students as a result of campus closures resulting from the pandemic, which may have contributed to further increases in nicotine
